RAPD Marker Related to Elongated Fruit Shape in Pepper (Capsicum annuum L.)

Abstract
Breeders develop pepper lines with different fruit shapes according to demands of market. It will be useful to develop an efficient tool for identification of fruit shape in pepper seedling. This could shorten a selection period from crossing to obtaining F-6 pure lines. We identified RAPD marker OPB-05-3 closely related with elongated fruit shape type in pepper. Experiments were carried out after crosses between two mother lines with semi-elongated fruit shape and two pollen donor cultivars, Palanacka babura and Palanacka kapija with elongated and bell fruit shape, respectively. Twenty primers with 9-12 bases were designed for RAPD, according to available sequence from NCBI database. Only primer OPB-05 with sequence TGCGCCCTTC amplified the similar band in cv. Palanacka kapija and all its F-1 hybrids. PCR product was reproducible, but the marker was always a weak band. Primer sequence and PCR conditions will be improve in the next period.
