AB  - Agrobacterium rhizogenes A4M70GUS-mediated transformation of Savoy cabbage (Brassica oleracea L. var. sabauda) and two local lines of cabbage (B. oleracea L. var. capitata) was obtained using hypocotyl and cotyledon explants. The percentage of explants which formed roots was very high in all genotypes: 92.3 % in Savoy Gg-1, 64.4 % in cabbage P22I5, and 87.2 % in P34I5. Spontaneous shoot regeneration of excised root cultures grown on the hormone-free medium occurred in all three genotypes. In cabbage lines P22I5 and P34I5 shoot regeneration was higher (9.3 and 2.6 % respectively) than in Savoy cabbage Gg-1 (1.3 %). Transgenic nature of hairy root-derived plants was evaluated by GUS histological test and PCR analysis. All the tested cabbage shoots were GUS positive whilst in a Savoy cabbage GUS expression was registered only in 55 % of tested clones. PCR analysis demonstrated the presence of the GUS gene in regenerated shoot clones and in T-1 progeny.

Introduction of resistance to herbicide Basta (R) in Savoy cabbage

AB  - Resistance to herbicide Basta(R) was introduced into pure inbred lines of Savoy cabbage ( Brassica oleracea L. var. sabauda) by cocultivation of cotyledon and hypocotyl explants with Agrobacterium tumefaciens strains AGL1/pDM805 and LBA4404/pGKB5 (LB5-1). Shoot regeneration occurred on Murashige and Skoog medium supplemented with 1 mg dm(-3) 6-benzyladenine and 0.5 mg dm(-3) indole-3-butyric acid at 42.3 % and 71.4 % of hypocotyl explants treated with AGL1/pDM805 and LB5-1, respectively. Putative transformants that survived selection on 10 mg dm(-3) phosphinothricin (L-PPT) supplemented medium were confirmed by GUS assay and PCR analysis. The transformation rate was 58 % with AGL1/pDM805 and 25 % with LB5-1. Rooted plantlets were acclimated and then again screened for Basta(R)-resistance by spraying with 15 - 60 mg dm(-3) L-PPT. Surviving plants were selfed and Basta(R)- resistance was demonstrated in T-1 progeny.

Epistatic gene effects on the fruit shape of the parents of F1, F2, BC1 and BC2 progeny

AB  - We investigated the shape of tomato fruit by evaluating the ratio between the horizontal and vertical diameter in diallele crossings of 6 parental genotypes (D-150, S-49, S-35 and H-52, Kg-z and SP-109). The diallele and their progeny (F1, F2, BC1 and BC2), were used to investigate the mode of inheritance by generation mean analysis (Mather and Jinks, 1982). Additive gene effects were the most significant in affecting the inheriting the fruit shape. Most frequently expressed epistatic effects were the additive x dominance interaction, followed by additive x additive gene effects. The duplicate type of epistasis was relatively stable.

Afila gene effects on total yield of dry pea seed

AB  - The of gene at its recessive homozygous stage in peas causes the modification of leaves so that they only form vines with stipules instead of leaves of normal structure. The variety Filigreen of the afila type was crossed with twelve varieties with normal leaf structure. The aim was to produce afila genotype effects on the total yield of mature seed in their progeny. The genetic analysis of F1 and F2 generations (the segregation ratio between the normal and afila leaf type being 3:1) proved the monogenic character of this feature. The yield of mature seed in F1 generation, characterized by normal leaf structure, was higher than in the parents of both pea types. The total yield in dry seeds of the plants with normal leaf structure in F2 generation was 3% over the parents, whereas it was 6%, compared to the plants of F1 generation. The average yield of mature seed in afila pea plants was 4% lower than that of their parents, while it was 11% lower compared to the F1 generation. The decrease of the total yield of dry pea seed in F2 generation of the afila genotypes was conditioned by the reduction of the assimilation surface of the plant.

In vitro culture as a tool for improvement of cabbage cultivars in Yugoslavia

AB  - Cabbage (Brassica oleracea var. capitata) is one of the most common vegetables in Yugoslavia. Our breeding programs on Brassicas are developed with the aim to create genetically pure lines which could be used for creation of various hybrids and new cultivars. Since there are barriers which prevent fast realization of these goals, we have introduced in vitro culture techniques in our breeding programs. Thus we investigated possibilities of using micropropagation, anther culture, in vitro selection and genetic engineering methods as tools to improve our Brassicas and create new perspective genotypes suitable for uses in conventional breeding methods. So far we have clonned a number of perspective Brassica genotypes and identified the most appropriate medium for their growth and multiplication. We present here our efforts to overcome incompatibility barriers by attempting to create double-haploid genotypes of cabbage through anther culture. Identifying possible tolerance of our domestic populations to herbicide by in vitro selection is also one of our goals. We have also created our first transformed Brassicas, performing Agrobacterium-mediated transformation. In this paper we have presented our research in these areas.

We present here our efforts to overcome incompatibility barriers by attempting to create double-haploid genotypes of cabbage through anther culture. Identifying possible tolerance of our domestic populations to herbicide by in vitro selection is also one of our goals. We have also created our first transformed Brassicas, performing Agrobacterium-mediated transformation. In this paper we have presented our research in these areas.",

AB  - The correlation between seven quantitative features of tomato (yield per plant, fruit weight, fruit length and width, number of fruits per plant, pericarp thickness, and number of locules per fruit) was studied in seven parental genotypes and 21 F1 hybrids. In both investigated generations there was a significant positive correlation between yield per plant, fruit weight, fruit length and width, and pericarp thickness. A significant negative correlation was recorded between mean fruit mass and number of fruits per plant. A significant positive correlation was found between fruit length and fruit width. Both traits were positively correlated with mean fruit mass. The number of locules per fruit was significantly and positively correlated with fruit weight, fruit length, fruit width and number of fruits per plant, whereas no relation of yield and fruit pericarp thickness with the number of locules was recorded.

AB  - Our investigation was based on viewing the problem of inheriting anthocyanin and vitamin C contents in F-1 progeny of lettuce. It was presumed that progenies with increased contents of these substances could be obtained. Diallel crossing of eight lettuce genotypes of different anthocyanin and vitamin C contents were performed. Parental and F-1 generations were investigated comparatively, and their mode of inheritance was determined. Concerning the inheritance of anthocyanin, dominant genes prevailed and a higher content of this substance was succeeded in F-1 generation, whereas concerning the inheritance of vitamin C content, it was recorded dominance mode of inheritance, when compared to the parents with lower vitamin C content. Apart from the dominance mode of inheritance, significant additive gene effects in inheriting vitamin C content were recorded.

AB  - With the aim to improve the breeding process of cabbage (Brassica oleracea var. capitata) and to develop new cultivars in a shorter period of time, as well as to develop a basis for use of biotechnology methods, the in vitro propagation of Brassicas, was investigated. The experiments included 39 genotypes of different Brassicas, most of them cabbages. Shoot culture was obtained from segments of aseptically germinated seeds and from lateral shoots from plants grown in the open field and under cover. Shoot culture media for optimal growth and multiplication were elaborated. "Microplants" were planted in the open field. All the important conditions for the cloning process were evaluated.

AB  - Kupusnjače (Brassicaceae) su ekonomski veoma značajna familija biljaka. U poslednjih trideset godina jako mnogo se radi na poboljšavanju karakteristika vrsta roda Brassica. Medu novijim metodama koje se koriste u oplemenjivačke svrhe na kupusima jesu i tehnike genetičkog inženjerstva. Cilj ovog rada bio je da razmotri transformacije kupusnjača, prikaže preliminarne rezultate dobijem u našoj laboratoriji, gde je izvršena inokulacija nekoliko genotipova roda Brassica. Korišćenje soj A. tumefaciens koji nosi bar gen za rezistenciju na herbicid Basta. kao i gen za rezistenciju na kanamicin (nptII) i reporter gen za-glukuronidazu (GUS). U radu je dat i osvrt na svrsishodnost transformacija Brassicacea, kao i na eventualne rizike koje korišćenje transgenih kupusa nosi.
AB  - Brassicaceae are economically very important crop family. Improvement of its characteristics is in focus for the last thirty years very much. One of the methods of biotechnology used in Brassica genus is genetic transfor­mation. In this paper we have discussed transformations of Brassica genus, comparing it with the results that we achieved in our laboratory. We have performed inoculation with Agrobacterium tumefacien, with the construct containing resistance to herbicide Basta (bar), marker genes for Kanamycin resistance (npt II) and reporter gene-glucuronidase. We have also discussed risks of transgenic crop use and seed development.

AB  - Savremeni programi selekcije paradajza imaju za cilj stvaranje genotipova sa čvrstim plodovima. Takvi plodovi se tokom berbe i transporta manje oštećuju, čime se direktno utiče na njihovu veću upotrebnu vrednost. Ukrštanjem sedam divergentnih genotipova paradajza koji su se međusobno razlikovali po čvrstini ploda, metodom punog dialela bespovratnih ukrštanja. dobijen je 21 hibrid F1 generacije. Analizom komponenata genetičke varijanse ustanovljeno je da su dominantni geni imali preovlađujuću ulogu u nasleđivanju ovog svojstva. Posmatrajući sve kombinacije ukrštanja zajedno može se zaključili da je super dominacija predstavljala način nasleđivanja u F1 generaciji. Hibridna kombinacija dobijena ukrštanjem dva najbolja opšta kombinatora (V-100 X no-10) odlikovala se najboljom posebnom kombinacionom sposobnošću.
AB  - Present day program for tomato selection are aimed at creating the genotypes with firm fruit. The fruits with this quality surfer from minor injuries while being harvested and transported, which directly affects their better consumption purpose. By crossing seven divergent tomato genotypes that differed among themselves in fruit firmness, and by applying the method of full diallel without reciprocal crossings, we obtained 21 hybrids of F1 generation. Upon analyzing the components of the genetic variance we found out that dominant genes prevailed in inheriting this feature. Considering all the crossing combinations together, it could be concluded that super dominance was the mode of inheritance recorded in Fl generation. The hybrid combination obtained by crossing the two hybrids with the best general combining ability (V-100 x No-10) was characterized by the best specific combining ability.

Ukrštanjem sedam divergentnih genotipova paradajza koji su se međusobno razlikovali po čvrstini ploda, metodom punog dialela bespovratnih ukrštanja. dobijen je 21 hibrid F1 generacije. Analizom komponenata genetičke varijanse ustanovljeno je da su dominantni geni imali preovlađujuću ulogu u nasleđivanju ovog svojstva. Posmatrajući sve kombinacije ukrštanja zajedno može se zaključili da je super dominacija predstavljala način nasleđivanja u F1 generaciji. Hibridna kombinacija dobijena ukrštanjem dva najbolja opšta kombinatora (V-100 X no-10) odlikovala se najboljom posebnom kombinacionom sposobnošću., Present day program for tomato selection are aimed at creating the genotypes with firm fruit. The fruits with this quality surfer from minor injuries while being harvested and transported, which directly affects their better consumption purpose. By crossing seven divergent tomato genotypes that differed among themselves in fruit firmness, and by applying the method of full diallel without reciprocal crossings, we obtained 21 hybrids of F1 generation. Upon analyzing the components of the genetic variance we found out that dominant genes prevailed in inheriting this feature. Considering all the crossing combinations together, it could be concluded that super dominance was the mode of inheritance recorded in Fl generation. The hybrid combination obtained by crossing the two hybrids with the best general combining ability (V-100 x No-10) was characterized by the best specific combining ability.",

AB  - We performed the genetic analysis of 6 tomato hybrids originating from 6 inbred lines (D-150, S-49, S-35, H-52, Kg-z and SP-109). We produced their descendant generation F1, F2, BC1 and BC 2 In order to estimate the genetic effects, applied the additive-dominance model with three and six parameters. The epistatic effects were evaluated by using 6-parameter model (Mather and Jinks, 1982). There were significant differences between the mean values of the parents and their descendants concerning the number of fruits per flower branch. According to the results obtained from the investigated genetic parameters, the additive gene effects were prevailing for the feature of inheritance of the number of fruits per plant. We recorded the significant effects of these genes in all the investigated combinations. The estimated values of the epistatic gene effects additive x additive genes represented the only type of interallel interaction, which was found in the investigated crosses.

The effects of paraquat on cabbage (Brassica oleracea var. capitata L.) cultured in vitro

AB  - Considering the great toxicity paraquat has to a wide range of plant and animal species, including man, we have investigated the possible use of cabbage in vitro cultures as a model system suitable to study this pesticide. The main aim of this study was to investigate the effect of paraquat on growth and development of cabbage shoot cultures. Special attention was paid to the detection of eventual means by which cultures can overcome the detrimental effect of the chemical, and also to investigate if some of the genotypes coming from the breeding program have an increased tolerance to paraquat. Promising domestic genotypes PI94, P295, P6p, P44 were investigated in the rooting stage. A root induction medium containing MS inorganic salts and 5.0 mgl-1 IB A was also supplemented with Paraquat at 0.5, 2.0, 10.0, 50.0 mgl -1. Shoots were treated for 24 hours and then transferred to a hormone free medium. After 28 days we measured shoot length, number of internodes, percentage of rooting, root length and formation of laterals. Paraquat decreased all the parameters which were followed. On leaves it induced lesions. Genotypes P44 and P6p were more tolerant to paraquat then the other two genotypes. Genotype P44 was interesting since in some plants lateral shoot formation occurred.

AB  - In order to create Eliksir F1 a new hybrid, we performed a crossing of original sweet corn lines which were obtained from an artificial population by employing the inbreeding method. The aim of this paper is to compare the most significant characteristics of this hybrid and the following American hybrids: Sundance F1 Early Arctic F1, Spring Gold F1 and Reliance F1. The characteristics taken into consideration were: the earliness, total yield and the grain cob ratio. The experiment was carried out by using the random block system in four replications. The hybrids' differences were tested by Fisher's variance analysis. We found that Eliksir F1 was an early hybrid of sweet corn with the average of 1.3 ears and 1.7 side branches per plant. Eliksir F1 had the highest yield comparing to the other hybrids. The yield ranged from 11,955 kg/ha. The grain percentage was 62% of the total yield. Therefore, Eliksir F1 could be recommended as suitable for sweet corn hybrid early production.
